2.Cytosol and nuclear estrogen (ERc, ERn) and progesterone (PRc, PRn) receptors were measured with the Dextran-coated Charcoal method in 50 ovarian tumors and 6 normal ovarian specimens.
5.1. Proliferation assay of MCF-7 human breast cancer cells: cells(2-3 x104 cells/ml) were seeded in culture plates of 96 wells. Each well contained 100ul RPMI-1640 medium(phenol red-free) supplemented with 10% dextran/Charcoal-treated fetal bovine serum (FBS) and insulin(30u/mL).
7.Estrogen receptors (ER) of the spayed rat uterus were measured by modified dextran coated charcoal assay(DCC) ,after injection of estradiol (E) or simultaneous injection of estradiol and tamoxifen (E+T) 6h to 64 days respectively. The level of ER in E group and E+T group were both increased with the extention of the time after the injection.
8.In 56 cases the PAP method was compared with the ABO (avidin-biotin-peroxidase complex) immuno-histoehemical method and DGO (dextran coated charcoal) biochemical assay in the same specimens. Comparison of the PAP and DCC methods showed a total correspondence rate of 71.4% and a correspondence rate of 86.8% in positive cases.
