conformation analysis
.Construction and space conformation analysis of the fusion gene of human IgG3 upper hinge region and p53 tetramerization domain
.In this paper, B3LYP/6-311G(d,p) method was applied to study the conformation stabilities of dipeptide molecule CH_3CONHCH_3. Conformation analysis was carried out from the point of view of intramolecule hydrogen bond interaction, steric interaction, and Coulomb interactions.
.The methylation patterns of 13 CG dinucleotide sites in CpG islands of BRCA1 promoter from normal blood DNA after treatment with methylase M SssI were analyzed by methylation sensitive single strand conformation analysis (MS SSCA) and DNA sequencing Eight different migration patterns observed by MS SSCA were confirmed as eight different methylation patterns by DNA sequencing These researches will provide reference for the study of BRCA1 gene methylation patterns in clinic breast cancer samples.
.An A/C polymorphism was identified at nucleotide-11 from the acceptor site of IVS3 of the phenylalanine hydroxylase gene. This polymorphism can be detected easily by means of single strand conformation polymorphism analysis.
.Expression of Fas was analyzed by a FACScan flow cytometry. Transcription and mutation of Fas gene were detected by RT PCR and single stranded conformation polymorphism(SSCP) analysis.
.Methods The study included 16 cases of PCG from different district of China. Direct sequencing evaluated the coding and the promoter regions of CYP1B1 in PCG patients. Single strand conformation polymorphism (SSCP) analysis was used in normal groups to screen other mutations.
.To discuss the molecuar mechanism of the small cell lung cancer (SCLC). P53 gene mutation in SCLC of 14 paraffin embedded tissue section was observed by immunohistochemical staining (LSAB method) and polymerase chain reaction and single-strand conformation polymorsphism (pcr-SSCP) analysis.
.Methods Polymerase chain reaction-single strand conformation polymor-phism (PCR-SSCP) analysis and direct sequencing were used to screen the whole Cx43 coding sequence in 37 patients with CTD, 26 patients with other heart defects and 10 normai children.
.Five organic compounds were selected and their structures were determined by X ray diffraction. It was found that the disordered arrangement (occupying rate, position) existing in the partial atoms under the crystal state leads to the exchange in the molecular conformation, and by means of X-ray diffraction analysis, the average molecular structures are obtained. The disorder phenomena in the organic crystal structures exist universally, which make it more difficult to determine the crystal structures correctly in three-dimensional space.
